I dropped a screwdriver and poked a hole in the rubber surroundings of
my 4x6 speaker.

This will obvious affect the sound.... since it cant move air to well
with a hole in it...

Any way i can patch up the hole???

I dont plan on cranking my speakers excessively loud.... so would a
hot glue gun dab of glue work?

Black silicone rtv should work fine and keep the flexability intact.
A hole will not affect the sound anyway, but its nice to fix up.

Oh, is it black rubber or is it foam? Regardless, rtv will work.

The hole will likely get bigger if it isn't fixed. Of course depending on
how hard the speaker is driven.

A lot of people use super glue. I can't speak from personal experience, I've
never poked a hole in one of my speakers =P.
